Genetic diversity and differentiation among populations of the Mecardonia acuminata (Plantaginaceae) complex inferred from inter simple sequence repeats [ISSR] banding patterns. MECARDONIA acuminata is a widespread wetland species in southeastern USA, distributed throughout the region. Previous classifications propose three subspecies based on the shape and size of leaves, and length of pedicel (subspecies acuminata, peninsularis and microphyla). Subspecies acuminata is widespread and occurs throughout most of the distributional range of the species, subspecies microphyla is found from southern Georgia to southeastern Louisiana, and subspecies peninsularis occurs only in southern Florida. We employed analysis of variation among inter simple sequence repeats (ISSR) to elucidate the genetic relationships among 22 populations of the M. acuminata complex sampled from 7 states ranging from Texas and Tennessee to Florida. Eighty four loci scored for seven primers were utilized in the genetic investigation of 233 individuals. PCR fragments sizes ranged from 200bp to 1600 bp. For each primer examined, 8 to 15 loci were scored, demonstrating high polymorphism of ISSR markers within species. The data obtained will be used to analyze genetic differentiation among populations, and to determine the genetic structure of the species complex. Levels of gene flow will be assessed among populations particularly sampled within ranges of overlap of subspecies. Results of the current study demonstrate the utility of ISSR markers in elucidating the level of genetic diversity within the M. acuminata complex. Log in to add this item to your schedule
